She is the eldest child of the late Rose Marie Cassaniti (1914 – 2000) and the late Danny Thomas (1912 – 1991). Her father was a professional comedian with the roots of Lebanese Roman Catholic while her mother was of Sicilian ethnicity. Thomas has two younger siblings, Terre Thomas and Tony Thomas, whereby Tony is a producer by profession. Together with her siblings, Thomas was raised in Beverly Hills, California where her parents nicknamed her “Margo”. The nicknamed would later change to “Marlo” due to her childhood mispronunciation of the same.

As for school, Marlo Thomas attended Marymount High School in Los Angeles after which she joined the University of Southern California where she pursued a Bachelor of Arts degree. She graduated from the institution with Honors in Arts in 1959.
